Is there any down side to me playing one player in both my offensive and defensive line-ups. He is my best player, so I want to get maximum use out of him. I won my first game 42-14 doing this.

The cons outweigh the pros here, imo. Playing a player on both sides of the ball will affect his stamina. Also you will only be able to gain BPOS with him on the defensive side if he plays both defense and offense. Training must also be considered because players for offense and defense will need different skill sets.

YOu didn't win because you did that. You won because your opponent played a bot defense, so all his players were randomly assigned to the defense.
As TFS says, it isn't worth it to play a player both ways.
